PROLIA  (DENOSUMAB):  Denosumab is used to treat bone loss (osteoporosis) in women who are at high risk for bone fracture after menopause. It is also used to treat bone loss in men who are at high risk for bone fracture. Osteoporosis causes bones to become thinner and break more easily. Your chance of developing osteoporosis increases after menopause (in women), as you age, if someone in your family has osteoporosis, or if you take corticosteroid medications (such as prednisone) for long periods. Denosumab is also used to treat bone loss in women who are at high risk for bone fracture while receiving certain treatments for breast cancer. Denosumab is also used to treat bone loss in men who are at high risk of bone fracture while receiving certain treatments for prostate cancer that has not spread to other parts of the body. This medication works by slowing bone loss to help maintain strong bones and reduce the risk of broken bones (fractures). Denosumab belongs to a class of drugs called monoclonal antibodies.  1  osteoporosis Shortness of breath, hypertension, neuralgia, throat constriction, bone pain in leg and hip, face numbness, nausea, vomiting, light headed, fainting, uncontrolled weight loss, fatigue, and feeling as though I was dying. Symptoms persisted for months. It has been 10 months since my last injection, and I am still not feeling well. The symptoms of weakness, fatigue, lack of energy, and light headedness started after 2 weeks of my first injection. Three months after the first injection, I fainted and had my first hospitalization from the side effects, but the cause was unknown. Because of increasing symptoms and severity of symptoms, I had to resign from my job and stop driving after taking Prolia. It has robbed me of my quality of life. The prescribing doctor, Judity Castillo, MD, did not acknowledge that my symptoms were from Prolia, although the package insert mentions many of my symptoms. Prescribing physicians desperately need to be educated about the risks of this drug. F 86 3 times

 1  To prevent osteoporosis I had 7 Spontaneous Rebound Vertebral Fractures within the 18 months after I stopped having the Prolia injections. Also started having very painful IBS. Never had that previously. I think this drug should be a very last resort. F 74 12 months

 1  Osteoporosis Bad very scaly and itchy rash 4 days after injection. Then pain in my knees, thigh, groin. Then shoulder and arm. On anti inflammatory tablets but didn’t help much. Had 2 injections and had no idea I would be in so much pain. Couldn sleep! My doctor didn’t think it from from injection but I am sure it was and I had no issues up till that time F 66 6 months

 1  Osteoporosis Muscle pain from jaws to arms, legs, chest, back. Trouble breathing. Weakness to where hardly walk. Took one shot which was to last six months. NO WAY will do another one. Took my life away and very slowly getting it back. F 68 1 months

 1  OSTEOPOROSIS Prolia made me a cripple within 3 weeks of having the infusion. Severe groin, back, leg, arm muscle pain. Could barely dress myself, had difficulty walking, could only sleep on my back. I could not lift my arms, bend my legs or walk without being bent over. This condition lasted a full six months, and now 9 months later I am almost close to my pre-Prolia state. I believe the Prolia has done permanent damage to my muscles. Although I do not have the severe pain I suffered with originally, my muscles are not and don't believe ever will be the same as before. They are tight, inflexible, and feel like they will tear if I overextend them. One of the worst results of taking Prolia was that my rheumatologist misdiagnosed me with an auto-immune disease called Polymyalgia, which I did not have. She adamantly refused to attribute my condition to Prolia, which it was in fact the result of. F 65 1 days

 2  Osteoporosis 1st shot - severe left hip pain and pain in arms & hands. Night sweats. 2nd shot pain in hip & back. 3rd- severe pain in both hands, right hip, back and numbness in arms at night and now very sensitive skin with rashes. This all comes and goes... My doc said no one else has any side effects! it took me until the 2nd shot to think symptons may be related to this drug. I was fine before the shots. I don't want to break a bone but I don't know what to do.... I don't like this drug at all! F 63 1.6 years

 1  osteoporosis 3rd injection - immediate severe allergic reaction: instant WELT at injection site - hives, face & eyes swelling, throat closure, dizzy - all symptoms relieved by quick injection of Prednisone & 1 wk. of pills (Prednisone). After one year on Prolia, broke a toe, due to a misstep from a ladder - was not hard enough to normally cause a broken bone. F 78 18 months

 1  Osteoperosis Teeth became loose after first injection. After second injection severe jaw pain. Severe Headaches, Bone pain to the point I can barely walk for more than 5 minutes or even lift my arms to brush my teeth or comb my hair. Extreme spine pain. 5 Kidney infections and I only have 1 kidney. Dry skin, Ear infection and Blurred vision. My primary has dismissed these symptoms as side effects from Prolia however my nephrologist has not. F 54 1 years

 1  Osteoporosis So many bad side effects. Infections, sinusitis, ruined my good immunity, had such a bad inner ear infection I had to have an ear tube placed. I had never had an ear infection as an adult and apparently only 1-2 as a child per my mom. I also have had hair loss and skin rashes. Very upset with the Docs that prescribe this awful drug without warning of the side effects... F 63 2 times

 4  History of fractures with no obvious reason Absolutely none 100% true- no side effects . Now retired nurse who started Prolia while working Bone density has improved. ++ heartburn with Actonel. First rib fracture from coughing at 24 yrs. Three separate rib fractures when waking up in past 15 yrs. Broken toe with absolutely no history of trauma. Mother had similar history.  1  Osteoporosis I had no side effects with 1st shot, but after taking the 2nd one watch out. I got wierd blisters between my fingers and I'm in pain like flu like symptoms, nasty headache. Also while watching tv got dizziness like I was going to faint. Hey what if I was driving. Bad drug, I hope I get though this because it's only 1 week after my 2nd shot. Not a happy camper! I should have done more research on this drug. Prolia has severe side effects. A patient should run for the hills when their rematologist prescribes Prolia. F 59 7 months

 1  Osteoporosis Small itchy bumps and hair loss. The first shot I had no reaction but after my second shot I developed small itchy bumps and noticed my hair was thinning to the point where the scalp is visible in places. My hair is thin to begin with so this is the end of this drug! I can only hope that the hair loss will be replaced when this drug leaves my body!! My doctors kept calling me about taking this drug after they reviewed my dexa scan and I finally agree after being told the side effects are rare and many of their patients never experience any. I will not be taking a third shot and the industry must do better developing a drug for bones with better results. F 62 10 months

 1  Osteoporosis First injection was 11/17. One month later started to experience pain in the buttocks, knee and legs. Lasted for months and spoke to doctor who didn’t think that was the cause of my discomfort. We delayed second shot to 11/18 as I was feeling better. One week later I started experiencing severe left leg bone and hip pain. It’s not a coincidence that both times I experienced these side effects right after prolia shot. The doctor is not convinced prolia is the problem so is sending me for various tests. I am still in pain and hopefully it will subside. I do not plan on taking another prolia shot. Give consideration to quality of life before agreeing to prolia. I plan on reporting to the FDA. F 72 1 years
