PROLIA  (DENOSUMAB):  Denosumab is used to treat bone loss (osteoporosis) in women who are at high risk for bone fracture after menopause. It is also used to treat bone loss in men who are at high risk for bone fracture. Osteoporosis causes bones to become thinner and break more easily. Your chance of developing osteoporosis increases after menopause (in women), as you age, if someone in your family has osteoporosis, or if you take corticosteroid medications (such as prednisone) for long periods. Denosumab is also used to treat bone loss in women who are at high risk for bone fracture while receiving certain treatments for breast cancer. Denosumab is also used to treat bone loss in men who are at high risk of bone fracture while receiving certain treatments for prostate cancer that has not spread to other parts of the body. This medication works by slowing bone loss to help maintain strong bones and reduce the risk of broken bones (fractures). Denosumab belongs to a class of drugs called monoclonal antibodies. It prevents certain cells in the body (osteoclasts) from breaking down bone.    (Sources: U.S. Centers for Medicare Services, FDA)

1  Osteoporosis The day after getting the Prolia injection I started having severe anxiety to the point where I almost couldn’t function. I had to take Seroquel just to get through the day. My osteoporosis is severe with a T-score of -3.9 and I had a previous fracture, but I don’t know if it’s worth it. I can’t take the other osteoporosis medications because I had hyperparathyroidism and had 3 glands removed so all I can take is Prolia. I feel stuck and I don’t know what to do. If anyone else has experienced severe anxiety after Prolia please email me. It doesn’t come up,as a side effect, but I am 100% sure it caused the anxiety. F 53 6 months

1  Osteopina The FDA and Amgen should be ashamed. This drug either should not have been approved or mandatory information given to the patient. This drug has sent me to the emergency room for shortness of breath, heart palpitations, headache, blurry vision, severe muscle and bone pain. I have been sick for a year. I sometimes feel like I am actually dying. I have one month left of this injection left in my body and I pray no permanent damage has been done. I belong to a group on Facebook about Prolia complaints and there are hundreds of sick people because of Prolia. I realize some life saving drugs can be harsh but osteopina is not. Seems like there is a rat race with the pharmaceutical companies to hurry and get their drugs approved but without proper investigations and studies. Never, ever again will I be poisoned by Prolia. I would rather break every bone in my body. If you are thinking of getting Prolia injections investigate and study and learn about the drug. Once you are injected and if you have side effects you will suffer for at least six months. F 50 1 years

1  osteoporosis The day after receiving a prolia shot, both my arms broke out in a fiery red itchy rash and later with horrible stinging. Rash and itching would subside in one area of my body and move to another. I was miserable and got very little sleep for 6 months. I canceled the second prolia shot which was due 6 months later. In the last few days before the second shot was due, the tormenting itching and stinging rash significantly subsided. After 6 months, no rash. The only other medicine I take is blood pressure medicine that I have been on for 5 years and am still taking. F 79 6 months

1  Osteoporosis Hair loss, fatigue, cough, acid reflux, joint pain in knees, vertigo, including a severe episode shortly after 3rd shot. It's infuriating to have a physician tell you that you are the only one when reporting side effects, especially since I have visited a number of sites where others on Prolia have reported identical experiences. And the manufacturer is not being honest, either. While I don't want to end up with crumbling bones, I feel like my quality of life has taken a nose dive since starting the drug. And it's extremely depressing to have my hair falling out. The DEXA scan after 2 shots showed improvement in my hips, but spine worsened. At this point, I am not convinced that the trade-off is worth it. F 75 3 times

1  Osteoporosis I developed a rash/hives a week after my 3rd injection. I also developed severe crippling leg cramps at night, every night on and off all night! I have also lost at least 3/4 of my hair and it is still falling out! I have sore gums, headaches and my vision is not as crisp and sharp as it was. My joints and bones ache and this has all limited my quality of life greatly! I wish I had never agreed to this drug!!! My 3rd and never again Prolia injection was the first of March 2018.  1  Osteopenia I have osteonecrosis of the jaw and have been extremely sick since November 2017. All the doctors agree that the ONJ was caused by Prolia. The length of time I took the drug was 2 1/2 years. One injection is given q 6 months. This is a dangerous drug that can ruin your life. Be wary. The manufacturer claims such side effects are rare. That is untrue. I have 453 FB friends who have either ONJ, fractured femurs, and/or terrible muscle pain and weakness. We have been poisoned. F 60 5 days

1  prevent bone loss due to cancer Received Prolia at same time as Arimidex was prescribed so hard to prove what is the cause of health issues. However, after reading side affects of Prolia, will follow dates of treatments and side affects more carefully. Since starting treatment, I have Been experiencing tingling, numbness and burning of hands and arms while sleeping. Wakes me up several times during night but not every night. One time pain was so bad, I thought I was having a heart attack and went to the hospital for treatment. Dr's. keep telling me I have a pinched never in my neck, that is causing numbness. Will monitor what happens in coming days/weeks as I just had my third treatment of Prolia and will then make a decision as to receiving any further treatment if symptoms persist or increase.  1  Osteoporosis Worsening back pain, nausea, bone pain in legs that come and go and joint pain. The bone pain was sharp, burning and shooting pain while sitting at my office desk. Never had that happen until after the injection. Also having severe muscle spasms especially in arms and back! Do not plan on taking second injection. Would rather take my chances with the osteoporosis!! 11 days

1  Osteoporsis This is my fourth prolia shot and by far this one is the worst. Three days after my shot my feet starting burning and my lower legs and hands go numb when I sleep and they ache. Both doctors I have seen say its not a side affect--but I am not convinced. Also broke out in a rash with little bumps--that is finally better after about three weeks--but never again will I take this shot. F 61 4 days

1  To treat osteopenia condition Had terrible experience with this drug. My doctor recommended it to treat my osteopenia condition as a preventive measure, more than 2 years ago. Within a few weeks of the first shot, I developed hives and what looked like insect bites. I had been swimming in the sea daily so I thought I was bitten by sea lice. Six months later, after the second Prolia shot, all hell broke loose. I developed severe skin problems, itchy all over, hives appeared in any and every part of my body and they turned into blisters that would break and bleed. Every morning, my pajama would have blood spots in many places. By the seventh month of the last Prolia shot, I had to take Blexten (a strong antihistamine) daily to manage the problem and sleep became almost impossible. I put ice patches all over me at night to get relief so that I could catch some sleep. Finally, by the eleventh month after the last shot, with the help of Blexten, the itchiness was finally more tolerable. I am now in my eleventh month since the last shot and the itchiness is not over. I cursed my own stupidity for trusting my doctor and not do my own due diligence before taking Prolia. My doctor was totally helpless when I reported the problem to her and was not helpful. She kept saying that Prolia has little to no side effects, but she could not explain or treat my condition. Before you consider taking Prolia, consider my case as a warning. M 71 1 years
