PROLIA  (DENOSUMAB):  Denosumab is used to treat bone loss (osteoporosis) in women who are at high risk for bone fracture after menopause. It is also used to treat bone loss in men who are at high risk for bone fracture. Osteoporosis causes bones to become thinner and break more easily. Your chance of developing osteoporosis increases after menopause (in women), as you age, if someone in your family has osteoporosis, or if you take corticosteroid medications (such as prednisone) for long periods. Denosumab is also used to treat bone loss in women who are at high risk for bone fracture while receiving certain treatments for breast cancer. Denosumab is also used to treat bone loss in men who are at high risk of bone fracture while receiving certain treatments for prostate cancer that has not spread to other parts of the body. This medication works by slowing bone loss to help maintain strong bones and reduce the risk of broken bones (fractures). Denosumab belongs to a class of drugs called monoclonal antibodies. It prevents certain cells in the body (osteoclasts) from breaking down bone.    (Sources: U.S. Centers for Medicare Services, FDA)

1  Osteopenia Difficulty breathing, tightness to gain a full breath resulting in pain around sternum and rib cage. Swelling or tightness in back of throat made swallowing less natural to the point where my body didn't engage the swallowing mechanism. These side effects were the most difficult to experience - lasting approx 3 weeks. Other side effects lasting a day or so include the following: numbness in right hand and fingers, sciatica in right thigh, burned sensation on the roof of my mouth to the back of my throat, fatigue, and light-headedness. Now, a month later I am experiencing sudden weight gain. I have had one injection of prolia. It is important that those describing side effects explain how long their symptoms have lasted. At this point, I am afraid to have a second injection, though my doctor feels that my body will experience less of a reaction. The inability to breath properly and not being able to catch my breath was frightening. The tightness in the back of my throat making it difficult to swallow was even more frightening, but this subsided more quickly than the breathing problem. F 66 1 times

1  Bone density Severe pain and muscle spasms in joints and muscles that begin every night around any time from 3am on. I try to walk it off but right now I am using a walker because I had knee surgery the same week that I had my 1st and only Prolia injection. It's been a nightmare. Also have pain between my shoulder blades. Bladder problems that I had to stay home for fear of accidents and have appointment with urologist later this month. This is only part of what I have experienced. Never take another Prolia injection. I feel like a recluse because I just get exhausted trying to go anywhere. This is not what I thought I would be doing with my life. Also there is brain fog-I have been embarrassed by forgetting a word in the middle of a sentence. Oh-I also have been recovering from hip surgery in May 2017. It is a not a F 69 1 times

1  Severe Osteoporosis With first shot had digestive issues and sores in mouth...2nd shot had inflamed tendons in both heels (I am an avid hiker and that was NOT a good thing!) and extreme muscle soreness after exercise. Also had a HORRIBLE case of Bronchitis that was very hard to get rid of. 3rd shot...insomnia, severe constipation and tightness and burning pain in back of neck and low back. I have Fibromyalgia and now THAT is flaring up! Back on Lyrica for pain and Benefiber for constipation...also depressed. Blood pressure is way up so will need medicine for THAT! Will NOT be getting a 4th shot. I will take my chances! Try it once ....see what happens. If you get reactions, like most of us don't get another shot no matter how convincing the Doctor is. We do need a QUALITY of life! F 67

1  Osteopenia I have osteonecrosis of the jaw and have been extremely sick since November 2017. All the doctors agree that the ONJ was caused by Prolia. The length of time I took the drug was 2 1/2 years. One injection is given q 6 months. This is a dangerous drug that can ruin your life. Be wary. The manufacturer claims such side effects are rare. That is untrue. I have 453 FB friends who have either ONJ, fractured femurs, and/or terrible muscle pain and weakness. We have been poisoned. F 60 5 days

1  prevent bone loss due to cancer Received Prolia at same time as Arimidex was prescribed so hard to prove what is the cause of health issues. However, after reading side affects of Prolia, will follow dates of treatments and side affects more carefully. Since starting treatment, I have Been experiencing tingling, numbness and burning of hands and arms while sleeping. Wakes me up several times during night but not every night. One time pain was so bad, I thought I was having a heart attack and went to the hospital for treatment. Dr's. keep telling me I have a pinched never in my neck, that is causing numbness. Will monitor what happens in coming days/weeks as I just had my third treatment of Prolia and will then make a decision as to receiving any further treatment if symptoms persist or increase.  1  osteopenia Please read this before taking the PROLIA injection. My endocrinologist recommended prolia for me after taking the drug called ACTONEL for 8 months. The Actonel's side effects were killing me, literally... After prolia was approved by my insurance company, the drug was shipped to my doctor. He specifically told me, because I asked, that the side effects from prolia were mild. He said the same thing about Actonel. I called his office today after doing extensive research on this drug and canceled my appointment for the injection. I hate to say it...but I think these doctors are receiving a kick back from the drug companies for having patients taking these horrible drugs. I'm going to continue taking vitamin "D" and calcium supplements and hope for the best. I'm very active and I feel great now after I stopped the actonel. I'll never get talked into a medical trap again.. M 63 0 days

1  Osteoporosis Bone and muscle pain. The bone and muscle pain is off the chart. My husband has had 1 shot in July 2016. He will not get another one. He was disabled before this shot but since getting it, he is worse. Prolia is suppose to help but he thinks it makes it worse and should be taken off the market. M 62 6 months

1  osteoparosis None I have been reading reviews about side effects, to determine if I want to use Prolia. It seems that the vast majority of people who use it with little side effect are younger, people in their 40's to early 60's. Older users seem to have the most side effects. I'm not willing to take the chance of negative side effects M 67 0 days

1  To treat osteopenia condition Had terrible experience with this drug. My doctor recommended it to treat my osteopenia condition as a preventive measure, more than 2 years ago. Within a few weeks of the first shot, I developed hives and what looked like insect bites. I had been swimming in the sea daily so I thought I was bitten by sea lice. Six months later, after the second Prolia shot, all hell broke loose. I developed severe skin problems, itchy all over, hives appeared in any and every part of my body and they turned into blisters that would break and bleed. Every morning, my pajama would have blood spots in many places. By the seventh month of the last Prolia shot, I had to take Blexten (a strong antihistamine) daily to manage the problem and sleep became almost impossible. I put ice patches all over me at night to get relief so that I could catch some sleep. Finally, by the eleventh month after the last shot, with the help of Blexten, the itchiness was finally more tolerable. I am now in my eleventh month since the last shot and the itchiness is not over. I cursed my own stupidity for trusting my doctor and not do my own due diligence before taking Prolia. My doctor was totally helpless when I reported the problem to her and was not helpful. She kept saying that Prolia has little to no side effects, but she could not explain or treat my condition. Before you consider taking Prolia, consider my case as a warning. M 71 1 years
