LUPRON DEPOT  (LEUPROLIDE ACETATE):  Leuprolide is used to treat advanced prostate cancer in men. It is not a cure. Most types of prostate cancer need the male hormone testosterone to grow and spread. Leuprolide works by reducing the amount of testosterone that the body makes. This helps slow or stop the growth of cancer cells and helps relieve symptoms such as painful/difficult urination. Talk to your doctor about the risks and benefits of treatment.   FDA Approval Date: 1989-01-26 (Sources: U.S. Centers for Medicare Services, FDA)

5  adenomyosis Hot flashes, weight gain. I went from having debilitating pain every day to having a twinge a few times a week. I was pain free for the first time in 6 years. However, I gained about 50 lbs in my 6 month treatment without changing any of my diet/exercise habits. F 23 6 months

4  Endometriosis Hot Flashes, mininmal moodyness I had surgery in December for stage 4 endo. I had one three month shot of lupron. I did not have any major side effects except for hot flashes. They were annoying but nothing that I could not live with. I have still not gotten my full period back yet and it is kinda of nice. Hope to try and get pregnant soon but was supposed to wait until I got my period again. Wondering if anyone who had the 3 month shot has had to wait longer than three months to get their period again? F 35 3 months

4  endo Hot flashes, night sweats, mood swings, sleeplessness, fatigue, back pain, vaginal dryness, memory loss. I'll get my 4th shot out of 6 next week. I'd say in the beginning, the first few weeks it made it all worse, which was expected due to the increase in estrogen. After a month the hot flashes started, but I didn't mind since my period was gone. With my period, I had horrible pain 3 weeks a month. The hot flashes have gotten unbearable. I sweat like a pig! My heart thumps during them. I haven't gained a pound, but I do exercise. All of a sudden though, I am in excruciating pain in my lower back and pelvis. Is anyone else experiencing this? I am going to call my ob/gyn tomorrow. Other than that, I have to admit I was terrified when reading how many people gained tons of weight. I have been overly concious of my eating pattern.  5  Endometriosis Hot Flashes and weight loss. Also a little spotting the first month. I have been dealing with endometriosis for almost 10 years and am on my second round of Lupron. The first time in 2001 I took it for 6 months and it worked great. A few hot flashes are nothing compared to the relief from pain. I was able to concieve within 6 months of completing it and have done well until lately. I have never been able to tolerate the pill and have had 7 or 8 laporoscopies. This is my last cycle of it. When this is done I'm having a hysterectomy. It works great but you can only use it for so long. I started my second round of it a few weeks ago and have just begun to feel like a normal person. Thank God for small miricles! F 28 3 days

2  Endometriosis Three days after the first injection I have a non stop headache and the pain that was told to me would go away hasn't it's gotten worse. I hope this goes away. I've already gained 30lbs from the endometriosis which in itself causes weight gain. I'm not sure if this medication will do what it was told to me it would do but I do know at 1300.00 an injection something better give. My doctor told me to take motrin 600mg which has me in the bathroom every five minutes. I honestly believe the doctors don't realize the pain associated with endo. My doctor told me it was probably all in my mind. But he didn't say that after He couldn't rmove the endo from around my bladder and other places.. He took pictures of the growths and they had begun to tear and I had internal bleeding which he had to cauterize. Some one tell me if I am imagining pain that wake me up in the middle of the night and bleeding and clotting so bad I don't feel like moving. I'm a mother of 3 and I can not continue to live my life like this. F 33 3 days

5  Endometriosis Hot flashes and night sweats. Some vaginal bleeding in first couple of weeks. Some memory loss, but it's negligible. I discovered that when I exercised (cardio + weight resistance) 3 times a week, the hot flashes were minimal. When I stopped exercising, the hot flashes came back in full force! Needless to say, I am back to exercising and after 2 weeks the hot flashes are far easier to deal with. I have not experienced any weight gain. It definitely is much less scarier than I thought it would be. F 41 6 months

3  Oestrogen sensitive Breast Cancer Vaginal dryness - total atrophy, especially the first time I took it. Nothing helped. Within a month, sex was impossible. Hot flushes, reduced concentration and brain-power (I am a full-time student of Pharmacy). Exhaustion, sleepiness, joint pains. Probably caused the osteoarthritis detected in my recent scan. Osteoporosis developed very quickly, but bones improved quickly when I went off the drug for a while. I now take Actonel (a bisphosphonate) to prevent bone loss. I developed metastases when I went off the drug for a while, including while I was in a gene therapy trial. Don't people in the States take this drug for breast cancer ? It really does work if your cancer is oestrogen sensitive - you take it if you are pre-menopausal, to shut down oestrogen production by the ovaries. Then you can - and probably need to - take Arimidex as well, to stop peripheral production of oestrogen (by the fat cells, adrenal glands, etc.) I need to take both, as the Prostap / Lupron alone doesn't totally control the cancer. By the way, in the UK, Lupron is called Prostap. I have been living with breast cancer since at least 1997, and have only had this anti-oestrogen therapy - not chemo, because I had hoped to have children and didn't want to damage my fertility. Have taken Lupron / Prostap on and off for periods of 1-2 years. F 48

2  endometriosis headaches, mood swings!!!!!, nausea/vomiting, still having cramping/pain, joint/bone pain, memory loss I was just diagnosed with endo on April, 27, 2005. I had most of the usual symptoms except for one strange one, my belly button would bleed in conjunction with my period. I am to take Lupron for 7 months total and don't know if I can make it. The only reason I am trying this is because I want to have another child. I have either been crying or been so aggitated that my poor husband is sometimes afraid to speak to me. I am prone to migraines anyways but I have consistently had a headache and and usually sick to my stomach. I hope this gets better. F 29 12 days

3  Prostate Cancer Weight gain. Incredible shortness of breath. Unsteadyness on my feet. Even with regular exercise and walking I still feel terrible. I tire easily with just the slightest exhertion. I'm also impotent and testicles have shrunken. Can tell when it's time for the next 4 month shot because I always start to feel better. Doctor wants to keep me on Lupron one more year. I still have problems with urination and getting up 4-5 times a night. M 68 1 years

1  Endometriosis While I was on Lupron I immediately experienced an extreme mood change. I became fatigued to the point I had to stop working. I no longer had energy and was constantly craving food. I also experienced tenderness in my breasts. My side effects from Lupron changed my entire personality to a very negative out look on life and my body. Lupron Depot is a very strong medication that should not be forced onto younger adults. After taking Lupron I now look negatively towards my body because of the way this medication has made me feel. I am extremely dissatisfied with this drug, and would not recommend it to ANYBODY. F 22 5 months

1  Endometriosis Serious permanent side effects. Memory loss, fibromyalgia, chronic fatique, huge weight gain, arthritus, herniated discs, neck damage, swelling/fluid retention, allergies so severe surgery was required and fluid drains out my ears all day, IBS, headaches, mania, suicidal depression, post traumatic stress, susceptible to catch germs-colds. These are all PERMANENT and I haven't had a shot of lupron for 8 years. If you want to gamble with your life then try Lupron, but I suggest your read the package insert first. I lost everything to this drug, my career, social life, I moved to try to improve my health. Here's what you need to know: Lupron causes a phenominon called F 49

4  Stave IV Endometriosis Hotflashes/Nightsweats I originally went into the hospital because they found a cyst on my right ovary. After the surgery (laproscopic) it was determined that I had severe endo and probably had it my whole life. It was decided at that time that I would do a 3 month course of Lupron followed up by a full abdominal hysterectomy. I just received my 3rd shot today, and the only side effect that I have experienced at all are hotflashes/nightsweats. I have a previous history of anxiety and depression. I also suffer from a weight problem, so after reading much of the feedback from websites I was very nervous about losing my mind and gaining tons of weight once on Lupron. I am pleased to be a positive light on the use of this medication, because I have found it very beneficial. In fact, I never realized how miserable I was until I began the injections and started feeling better. As I have never tried to get pregnant and have always lived my life with heavy periods lasting more than 10 days each month accompanied by severe pain during ovulation and terrible cramps throughout, I am so glad to be rid of all of my pain and the bloated feeling that I have always felt is gone. Even though I have been conscious of what I have been eating while on the medication, because I was nervous of gaining weight, I have actually lost weight (approx 15 pounds in 60 days) without eliminating an occasional piece of cake or dish of ice cream. My surgery is scheduled May 20th and I am turning 40 tomorrow and I am feeling like I am beginning a whole new pain free chapter in my life and can't wait to find out what the end of this journey has in store for my well being. I certainly don't want to discredit anyone's personal experience on this drug, but after almost making the decision NOT to take this medication because of what I read, I felt it incredibly important to add my positive experience. The hotflashes are frequent (15-20 per day), but certainly not debilitating and they don't last for more than 2-5 minutes. I have not suffered from any anxiety, headaches, weight gain, hair loss, or insomnia, all of which I feared if I took this medication. I also had a spinal fusion 14 years ago so I was nervous about the bone density of my fusion being deminished by the medication but my doctor said that the Lupron would not have any effect on the strengh or stability of the bones. I don't believe that I am a person with a high pain tolerance or a person who handles not feeling good well, however, from my personal experience, I FEEL GREAT! I decided to take the monthly injections so that if I had any adverse reactions I could have the choice to stop after each month. I expressed all of my fears and anxieties with both my OBGYN and my regular doctor. They both took the time to give me their professional opinions related to my concerns. They told me that the potential for side effects were possible, but that the majority of the people who take the medication, either for endomedtriosis, fibroids, or prostate cancer, had no serious adverse reactions beyond hotflashes and possibily some headaches. Every person has to make their own decision, but know that there is a person out there who is glad that they took LUPRON. F 39 3 months

4  Fibroids Feelings of warmth (not full-fledged I am currently on my 2nd round of Lupron. I took the first round in 2000 before having surgery to remove my fibroids. Lupron worked great!!  5  endometriosis I have had almost no side effects. No weight gain (but I am a dietitan and I exercise regularly--I HIGHLY RECOMMEND YOGA FOR HORMONAL BALANCE!!!), no vaginal dryness, no memory issues, no mood swings. I have had all 3 shots for a 7 month course of treatment. After the second shot I had dizzy spells for about 2 weeks and I felt more tired than usual, but that's it. I am taking norethindrone, which helps with hot flashes (i haven't had any). Surgery (lap) and Lupron have ended all my pain and I am so happy I did it. I'm so sorry to see so many people have had negative side effects. F 28 7 months

4  Stage IV endometriosis None so far (thank God). Mild nausea and lightheadedness first day after shot. ? weight gain. I've only been taking it a month now after endometriosis was discovered during my surgery to remove 20 (!!) fibroids. No major side effects -- no hot flashes, memory problems, etc. I think I'm gaining weight though; I walk 2.5 miles 3 to 4 days a week and do circuit training 3 to 4 days a week, and I have only lost 1 pound this month. I'm hoping it's the old F 38 3 weeks

1  Fibroids 24hours after 1st shot I felt like I was going to crawl out of my skin! I was irritable and depressed. Side effects during my 3 consecutive shots were: terrible night sweats, hot flashes, memory loss, joint & muscle pain. Approx. 2 months after my last shot I started my period. A week prior to starting my period I was having terrible pain in both wrists and trigger fringer (thumb). I have senice been diagnosed with De Quervains tendonitis in both hands! I have a cast on my left hand and a splint on my right. I am convinced this is a result of being on Lupron since the signs were present during lupron treatment, but got terribly worse 1 month after my last shot. Aside from a myomectomy, i have had no other surgeries or health problems. F 33 3 months
