LUPRON DEPOT-3  (LEUPROLIDE ACETATE):  Leuprolide is used to stop early puberty (precocious puberty) in children. It helps to delay sexual development (such as growth of the breasts/testicles) and the start of menstrual periods. It also helps slow down early bone growth to increase the likelihood of reaching normal adult height. Leuprolide works by decreasing the amount of sex hormones that a child's body makes (estrogen in girls, testosterone in boys).   FDA Approval Date: 1997-03-07 (Sources: U.S. Centers for Medicare Services, FDA)

1  Precocious puberty I had mood swings, massive weight gain, migraines, hot flashes, and I broke multiple bones. I had swelling and rash at injection site. This medication ruined my life. I would never recommend this to anyone. F 21 5 years

4  Endometriosis I took Lupron for 4 years off and on. I am 29 years old and was very concerned about infertility after having 2 lap's to remove adhesions and still experiencing pain. I happened to have a very good experience with Lupron but I do urge one to do tons of homework on this med and it's side effects prior to taking it. After 3 months on Lupron, I had lost 10lbs, was not experiencing as much pain, felt much less moody and unpredictable, and did not have a period through the whole course of my treatement. However, I also experienced hot flashes that were out of this world, occasional fatigue, as well as osteopenia (or bone loss) in my hips and knees (Osteopenia has since improved with regular weight bearing exercise and calcium supplements) I stopped taking Lupron about 3 months ago and luckily all my symptoms went with it. My cycle is beginning to return but is in no way "a regular monthly cycle." I am very sorry to hear about the awful effects this med has had on so many women. I have not read many positive ratings on this med and do feel very fortunate after reading all of yours. I, like most of you, did not do my homework and jumped on the Lupron wagon with my eyes wide shut. My 2 concerns were; 1. no more pain and unbearable periods but most importantly 2. Fertility protection. From what I have read these have been the concerns of most women contemplating Lupron therapy. We all must weigh out the risks and benefits of starting any new medication and it is always best to get more than one opinion especially if you do not fully trust your physician. Also, there are many holistic therapies that have been effective in treating the pain associated with endo, fibroids, as well as other repoductive disorders. My recommendation to anyone considering Lupron therapy would be to seek out alternative therapies before putting this drug (or any drug) into your body. F 29 4 years

4  Prostate Cancer Hot flashes, zero libido, atrophy, weight gain. I was diagnosed with PC in 1999. Went thru: External conformal radiation 4 years later psa=7.0 Cryosurgery= no help Lupron 12/2004= the usual side effects. psa=0.26 Stopped Lupron 12/2007 I saw an article about a UCLA Med Center trial where 4,000 advanced PC men were divided into two groups 2,000 continued on Lupron 2,000 stopped Lupron and toke one 8 oz. of Pomegranate Jucie/day 4 years (as i recall) the results were that their psa was the same in each group with some in the pom juice group lower. My urologist agreed to my plan to stop Lupron and start pom juice. I started pom juice Sept 2007. PSA was 0.0.26 Checked PSA March 2008 it is now 0.10. I use the Pomegranate Juice concentrate (Sam's Club $16.00 += for 32 oz)(one oz = 8 oz of juice) so this about $0.50/day. Costco carries it also. Also available online. All the Lupron effects are slowly going away. To see the UCLA trial report search Google for "UCLA prostate Cancer pomegranate j M 79 3 years

1  Ovarian Fibroid and Heavy Bleeding Severe weight gain, horrific hot flashes, mood swings which effected my everyday life, severe depression, joint pain and frequent, long lasting headaches. I was happy with this medication on my first go around, I used it for a little over a year and it did what it was suppose to do, decreased pain, stopped my bleeding. After losing insurance coverage I had to go off this medication for about 2 years during which the pain and bleeding returned worse than before. I was able to get back on the shot for about 1 1/2 years during which the side effects were awful. I continued the shot because of the fear of the pain in my pelvic region returning and the uncontrollable bleeding. I have been off the shot since Feb and all my old symptoms are returning. This time I do believe I will seek a surgical treatment versus Lupron Depot! F 30 1.5 years

4  Endometriosis, cyst, bleeding, pain Hot flashes, moody, sobbing, bone loss I have been on Lupron twice for once for a year (2005) then another time for 6 months (2003). I start again today or tommorow whenever the shot comes in the mail. Lupron was a life saver for me, I went from bleeding for months and extreme pain to almost no pain at all. This time the main reason is for a cyst on both ovaries. I am trying to avoid another surgery due to me still trying to conceive. F 31 1 years

4  Prostate cancer I am taking Lupron for two years for post op Prostate Cancer and removal of prostate. I had 33 radiation treatments followed by injection of Lupron ( 3rd one ) 3 more to go. I am getting severe pain in one knee with swelling and mild and occasional pain in the other knee. Some days are better than others and Advil seems to minimze pain and I walk normal. Did some kneeling during some construction and days later ... interior meniscus of knee had acute pain. Chiropractor seemed to alleviate some symptoms but next it has moved to outside of knee in meniscus and ligaments an causing to limp and be in pain when I walk. Is this caused by LUPRON ??  3  endometrosis hot flashes, cold chills, migraines, nausea, difficulty urinating, constipation, painful female organs, anxiety, overwhelmed, fatigue, acne, weight gain (40 lbs), memory loss, throbbing in my right temple that makes me tired--(and probably more side effects I can't remember right now) I gave this medicine a 3 because even though I had a blue million side effects, I had severe endo and my organs were stuck together. The doctor couldn't remove it because he didn't want to damage my other organs. So I did lupron once a month for 6 months. The first 5 months, I was great on the shot with only acne and hot flashes as side effects. The 6th month however brought with it the rest of the side effects. However, the doc scheduled my surgery in October of 2005 after my last shot in August and was able to clean me out from the endo. In November I went on the shot for another 6 months due to the pelvic pain I was in while on the last month of the first 6 shots and wow, did it ever get worse. I thought it was the endo causing the pain but little did I know it was the shot. During this time side effects were worse than ever and 5 years later, I am still suffering from them. F 34 1 years

1  Pain, endo? Had an emergency hysterectomy after the birth of my son 19 years ago, have had trouble ever since, it's taken them this long to do anything!!! passing out with the pain, only one ovary, adhesions, leg pain, bowel problems I was great at first, even though the mood swings were awful and I was very very tired (I have narcolepsy and it seemed to make my symptoms there better too) but started getting chest pain within about 8 months and my blood pressure shot up!!..and stayed there! Massive weight gain, my hair has thinned a great deal ( I have very very long hair so it has made a big impact) The hot flushes started to get unbearable, I honestly felt that I would literally drop dead of heat exhaustion, dramatic but true! Suicidal!!!Lost all my confidence, have become a recluse..cannot lose weight no matter what I do, I'm so tired. Developed a fistula so now a colostomy is on the cards, the doctors can't tell me if it's connected or not. I just feel like a lab rat frankly so I'm not having the shot anymore, Totally fed up and have no life now. The oestrogen add back was the worst point. I was literally barking at the moon and I don't feel that I was monitored enough by the doctors. My life has ground to an abrupt halt completely and all the tiredness, hair loss, depression, it's all been for nothing. I wish I'd never been put on it. I will never go back on this drug, ever! I'm better of with nothing but my own oestrogen, at least the symptoms were only monthly rather than all the time! I don't believe that the doctors know enough to prescribe this drug to women. I studied medicine myself when I was younger and my treatment over the years has just been absolute rubbish! F 42 1 years

1  endometriosis hot flashes, significant mood swings, weight gain, skin crawl I took one full 6 month cycle of Lupron Depot. A year later I began another cycle after my hysterectomy. I quit taking the shots after the 2nd month.. I will NEVER take this medication again. The side effects were too severe! My personality completely changed while taking this drug. My doctor wants me to take the drug again because I am still in so much pain. I told her that I would rather endure the pain twice a month than take the medication. That is how bad I think the side effects are. I would really consider the side effects before taking this medication. Also, if you choose to take the medication, ask for the one month shot so you can quit if it is too hard on you. F 38 1 years

5  prostate cancer physical & mental exhaustion, loss of stamina, loss of concentration, hot sweats, raised liver functions & mild nausea, high blood pressure, high glucose level, raised cholestrol; increased appetite early on and periodically, bone thinning (first posted below on 4/2/08) - now finished Tx: this drug is like being on mild chemo, especially after the first 12 mos. BUT it seems to have worked for me. each man experiences it differently, but i do think it is harder the younger the patient, based on my support group experience.exercise is a great mitigator - i missed very few days - and resistance ex. is v. important - google RoAnne Segal's JCO article, 12/8/08 ascopubs.org/cgi/content/abstract/JCO.2007.15.4963v1. feel free to e-mail me for info and support.  5  premenopausal breast cancer After 2 yrs in combination with tamoxifen: thin eyebrows, cramping in calves, no periods (great!), but finally some severe cramping in calves and pain/swelling in am in knuckles and soles of feet. But I would do this combo again - not bad at all. Effects may be just menopausal. Used Lupron with Tamoxifen for 2 years for hormonal treatment of breast cancer. I believe Tamoxifen does stimulate the endometrium because I got my period back after chemo, 1 month after starting tamoxifen. Therefore I used Lupron for ovarian suppression to hopefully keep my estrogen down and prevent reoccurance of my breast cancer. I think it was a good decision - no cancer return so far, and quality of life was OK. F 47 21 months

5  Endometriosis Positive Side Effects - no more migraines! Weight Loss - 40 pounds! Increased sex drive! Negative Side Effects - hot flashes, waking at night, but definitely worth it due to feeling SO much better! I took it for 7 months (no period for a year), then a year later took it again for 9 months (no period for a year again). I felt fantastic! No periods, no pain, could go to the gym and exercise, lost weight, wanted to have sex, migraines went away (did have regular headaches). I felt better those months than I had in a long, long time. However, when I stopped and my estrogen levels came back up, I had severe nausea for months - as if I were pregnant. F 43 16 months

1  Endometriosis Severe joint pain, hot flashes, insomnia, fatigue, low-grade fever, low sex drive, weight gain (from 112 to about 122), stabbing headaches, muscle weakness, vision changes. I started the Lupron injections six months after a failed laparoscopy to remove my endometriosis. For the first two injections I had the typical side effects of hot flashes, insomnia, fatigue, low-grade fever, and low sex drive. My third and final injection caused horrible joint pain that migrated around all of my joints aside for my spine and took a full year to ultimately dissipate. While taking the drug, I also had intermediate stabbing headaches that would come on in the back of my head, throbbing deeply and painfully with my heartbeat for a few minutes before disappearing. I have not had a recurrence of those headaches or the above symptoms per se. However, three years later, I now suffer easy tendon injury and flareups of joint pain as well as having multiple abnormally low hormone levels. I did ultimately decide on having a second laparoscopy where it was discovered that the majority of my pelvic pain was from a large ovarian cyst as well as multiple adhesions from my first surg F 30 9 months

In the end, Lupron didn't help me at all. I strongly suggest tryi F 30 9 months

4  Endometriosis-stage 3 weight gain 35 lbs., headaches, mood swings, hot flashes, loss of sex drive, memory loss, fatigue, insomnia, vaginal dryness, vision problems Depot Lupron was my only option other than having a hysterectomy to relieve the Endo pain and to also preserve my uterus. I haven't had children due to the Endo and my doctor hoped that if the Endo could be eradicated from my system that maybe I would have a chance to concieve. My doctor also said that at 43 I was about 10 years away from going through Menopause and going through it 'naturally' is better on a body then forcing it to go through it because of a hysterectomy. So with my doctor's reccommendation I went with the Depot Lupron. I received four injections;Aug 2004, Nov 2004, Jan 2005 and Apr 2005. In Jan 2005 my doctor put me on Progestin to relieve the severity of the HOT flashes that I was getting every-hour-on-the-hour, 24 hours a day, with each episode lasting approximately 15". I was miserable and extremely tired from a lack of quality sleep. By Dec 2004 my weight had started increasing and after starting the Progestin my weight rapidily increased.  3  severe and extensive bleeding from night swests, mood swings -- depression, joint pain I have taken three of the three-month shots and am scheduled for a fourth at the end of August. I WILL NOT be taking any more of this medication due to the increase of side effects. It seems that with every new injection, a new side effect rears its ugly head. At first, I as grateful since my periods stopped -- I had had severe and extended (sometimes six weeks) periods which were beginning to debilitate my life. The OB-GYN had tried every possible medication to help me, and she finally suggested Lupron Depot-3. At first. I was grateful for the cessation of my periods despite the night sweats and insomnia I experienced. With second shotcame the joint pain -- primarily in my fingers. With the third shot came the knee pain -- so painful that I have a problem getting up off the floor. My fingers are now so swollen in the morning that it is painful to bend them. While the medication didwhat it was supposed to do, the side effects render it difficult to continue. F 52 8 months
