LUPRON DEPOT-3  (LEUPROLIDE ACETATE):  Leuprolide is used to stop early puberty (precocious puberty) in children. It helps to delay sexual development (such as growth of the breasts/testicles) and the start of menstrual periods. It also helps slow down early bone growth to increase the likelihood of reaching normal adult height. Leuprolide works by decreasing the amount of sex hormones that a child's body makes (estrogen in girls, testosterone in boys).   FDA Approval Date: 1997-03-07 (Sources: U.S. Centers for Medicare Services, FDA)

4  Prostate Cancer Hot flashes, zero libido, atrophy, weight gain. I was diagnosed with PC in 1999. Went thru: External conformal radiation 4 years later psa=7.0 Cryosurgery= no help Lupron 12/2004= the usual side effects. psa=0.26 Stopped Lupron 12/2007 I saw an article about a UCLA Med Center trial where 4,000 advanced PC men were divided into two groups 2,000 continued on Lupron 2,000 stopped Lupron and toke one 8 oz. of Pomegranate Jucie/day 4 years (as i recall) the results were that their psa was the same in each group with some in the pom juice group lower. My urologist agreed to my plan to stop Lupron and start pom juice. I started pom juice Sept 2007. PSA was 0.0.26 Checked PSA March 2008 it is now 0.10. I use the Pomegranate Juice concentrate (Sam's Club $16.00 += for 32 oz)(one oz = 8 oz of juice) so this about $0.50/day. Costco carries it also. Also available online. All the Lupron effects are slowly going away. To see the UCLA trial report search Google for "UCLA prostate Cancer pomegranate j M 79 3 years

4  Prostate cancer I was prescribed this, which they call Prostap in the UK, as a preliminary treatment before having radiotherapy. After a few days I started to experience obsessions with certain things, and this developed into a mania over about 3 weeks. Then I had what felt like an emotional crisis, with insomnia, anxiety, extreme swings of mood (elation, weeping) and over-reaction to any emotional event. I felt I was deeply in love with one of my best friends! (Never told her) Also physical symptoms like palpitations and sweaty episodes, but these were insignificant in comparison to the mind-bending effects. After 3 months the effects are wearing off, thank god, And I am much calmer. Now I just have the effects of the radiation to suffer. It had the desired effect on my prostate cancer, but I hope I never have to have this drug on a long-term basis. M 65 3 months

1  prostae cancer Hot flashes, extreme sensitivity to sun. Mania (without depression) Loss of libido. Difficulty with erections. Orgasm without ejaculation. Took 2 one-months shots. No problem. Then one three-month shot: after two months 24-hour hot flashes and recurrent bouts of mania. I don't mind about the loss of libido. I expected that. Shot was supposed to have worn off one month ago, but the effects are worse than ever, while my psa slowly climbs. I'll never have any kind of treatment for prostate cancer again. I'm not afraid to die if I have to live like this. M 64 5 months

5  prostate cancer physical & mental exhaustion, loss of stamina, loss of concentration, hot sweats, raised liver functions & mild nausea, high blood pressure, high glucose level, raised cholestrol; increased appetite early on and periodically, bone thinning (first posted below on 4/2/08) - now finished Tx: this drug is like being on mild chemo, especially after the first 12 mos. BUT it seems to have worked for me. each man experiences it differently, but i do think it is harder the younger the patient, based on my support group experience.exercise is a great mitigator - i missed very few days - and resistance ex. is v. important - google RoAnne Segal's JCO article, 12/8/08 ascopubs.org/cgi/content/abstract/JCO.2007.15.4963v1. feel free to e-mail me for info and support.  4  Prostate Cancer I have been on Lupron Depot 3 for 4 months, I had prostectomy July 2007, The only side effects I have had from Lupron are hot flashes and lower back ache. both of which are tollerable, I was very wary at having Lupron injection , I read alot of the comments on this site plus many other sites, But I must admit I really have not had any major side effects from this drug, I feel it is everyones choice as to whether you have this drug injected, Everyone reacts differently to all chemicals , I feel the positives out weigh the negatives with this drug, I suggest people should use their own discretion in decisions, Not to just take what others have to say, We are all indaviduals and God gave us the ability to think for ourselves, Use that ability in every way possible to fight what ever cancer or disease you may have. I attend a support group each month, I have found it a great way to learn, It also allows you to be able to speak about your experiances regarding Lupron , Radiation ECT. I can only speak of my experiance with this drug, Maybe it works better for males than females, But I feel one never knows unless you try it, But please dont believe all the negatives you re M 59 120 days

1  PROSTATE CANCER In 2007 I had a radical prostatectomy. In 2009 my PSA went up. Dr recommended Lupron depot. Did not inform me of possible side effects. Now I have no muscle strength, very poor endurance, hot flashes, very poor sleep, memory loss, very poor concentration, irritability, bouts of extreme depression, vision changes, and PAIN in legs. When I went back to Dr he said my PSA was lower and would not need another injection. Stated I would never take another injection. This medicine has destroyed my life. I have been a paraplegic for 23 years. My life has gone from being independent to depending on others. I cannot make transfer into my car so now will have to buy van with lift. The pain in my legs seems to be bone pain mainly is in my left femur which has had no feeling for 20+ years. Due to the muscle weakness I have fallen and fractured two bones in calf. Dislocated a finger and suffered a torn rotator cuff and head trauma. THIS MEDICINE SHOULD NEVER BE GIVEN TO A PARAPLEGIC WITHOUT AN EXPLANATION OF POSSIBLE SIDE EFFECTS! I was given the shot Sept 1, 2009 and went to my 40th class reunion on Nov 21, 2009 and barely remember any of it. M 59 1 times

2  prostate cancer (T3a, 4+4) hot sweats, raised liver functions, high blood pressure, increased appetite, loss of stamina, bone thinning (first entered under 'lupron' but actually receiving L depot-3) typical hot sweats that seem to be decreasing with time - good since i have a 28-month program. ALT, AST and cholestrol were very high initially and have returned to top of normal; sugar higher and B.P. a problem too; also i'm always hungry. i've always had an aggressive daily exercise routine so i've confined weight gain to around 5# and believe exercise is key to L. management. it is certainly a little tougher to scull or cycle, but my yellow wristband is an inspiration. exercise also helps fight bone thinning - i learnt i had borderline osteoporosis readings in my spine from the baseline bone density scan. is anyone on a prescribed course other than 4-6 months and 28 months initially? i am concerned about becoming hormone refractory. i've been on effexor for 7 years for depression (strong genetic history) so can't tell whether it's helping with the sweats/mindset. PSA has declined from 13.1 to 0.1, so clearly the L is d M 57 5 months

4  Prostate cancer I am taking Lupron for two years for post op Prostate Cancer and removal of prostate. I had 33 radiation treatments followed by injection of Lupron ( 3rd one ) 3 more to go. I am getting severe pain in one knee with swelling and mild and occasional pain in the other knee. Some days are better than others and Advil seems to minimze pain and I walk normal. Did some kneeling during some construction and days later ... interior meniscus of knee had acute pain. Chiropractor seemed to alleviate some symptoms but next it has moved to outside of knee in meniscus and ligaments an causing to limp and be in pain when I walk. Is this caused by LUPRON ??  3  severe and extensive bleeding from night swests, mood swings -- depression, joint pain I have taken three of the three-month shots and am scheduled for a fourth at the end of August. I WILL NOT be taking any more of this medication due to the increase of side effects. It seems that with every new injection, a new side effect rears its ugly head. At first, I as grateful since my periods stopped -- I had had severe and extended (sometimes six weeks) periods which were beginning to debilitate my life. The OB-GYN had tried every possible medication to help me, and she finally suggested Lupron Depot-3. At first. I was grateful for the cessation of my periods despite the night sweats and insomnia I experienced. With second shotcame the joint pain -- primarily in my fingers. With the third shot came the knee pain -- so painful that I have a problem getting up off the floor. My fingers are now so swollen in the morning that it is painful to bend them. While the medication didwhat it was supposed to do, the side effects render it difficult to continue. F 52 8 months

3  Large Fibroid 10.5cm,in uterus/cerv Wake up with Headache, stiff back n ribs, flu like symptoms, feel extra tried, BUT NO mood swings or hot flushes yet. Abdomen increased in size after 1 week and was very uncomfortable, peeing every hour. By week 2, some decrease in abdomen but bleeding around normal period time. Treatment is to shrink fibroid prior to surgery, currently abdominal hysterectomy booked for July, desperately want to avoid surgery or want option for sub total not total hysterectomy. Tried Marina for six months prior but fibroid continued to grow. Hope this works as out of options. F 50 14 days

Memory loss/Forgetful Initially agreed to to have 3 x monthly PROSTAP injections, prior to my hysterectomy. Ended up having 5 as they couldn't fit me in for the op.  5  ovarian cysts and uterine fibroids insomnia first week, mild hot flashes, mild nausea For me this is a miracle drug. It shrank the cysts and fibroids in 3 months and I was able to avoid surgery to remove my uterus and ovaries. I REALLY wanted to avoid the surgery--my mother contracted a lung infection during her hysterectomy and never really recovered from it. But my grandmother died of ovarian cancer and I was scared. In my situation, the side effects are so minor compared to the alternatives. F 49 4 months

3  Premenopausal breast cancer Weight gain, loss of energy, dyspareunia, depression. I have taken this for estrogen sensitive breast cancer - to put me into a premature artifical menopause, so that I can take the anti-hormonal medications usually given to postmenopausal women. Lupron is called Prostap in the UK, and the 3-month injection has the same effects as the once-a-month injection, but (obviously) with only one injection every three months. Am now back on the once-a-month injections, this time with fulvestrant (faslodex) instead of arimidex, which stopped working. If the fulvestrant works, I might go back on the once-every-3-months injections. Otherwise, I shall go off both and have to undergo chemotherapy. Side effects include weight gain (including middle-age spread), tiredness, dry hair (?), vaginal atrophy (dyspareunia), etc. plus, not surprisingly, depression. F 49 6 months

2  Uterine fibroids Briefly? Leg pain, vertigo, nausea, vomiting, fever, chills, sweats, flu-like symptoms, debilitation, fatigue I have known I've had fibroids for some 15 years now, but they (one 15cm one, plus other smaller ones) have become so problematic that my uterus was the size of a six-month pregnancy, and was pressing on my leg veins (I've had a clot), and central veins (feeling dizzy if I lay on my back for too long). I am of course a prime candidate for hysterectomy, but do not want to have one for various reasons, so was keen to try Prostap (UK name). Was given the treatment (1-month depot injection) by the nurse who said that most women are fine, without being told of the possible adverse side effects. It was only after reading the leaflet later, that I became concerned. Felt OK in the first 2 weeks of treatment, and it was like a miracle in terms of the notable uterine shrinkage. My stomach became flat again, and my legs stopped swelling up. Great! However, I then developed the excrutiating leg pain (bone pain), that kept me awak at night. Next was the vertigo attacks. Usually on rising in the morning, but so bad that I had to take time off work. Next was the worse nausea and vomiting I have experienced in my life, to the point I had to go to A&E with dehydration from it. Then came the flu-like symptoms and the hot flushes, plus the affects on sleep. As a business woman and fitness enthusiast, the treatment has totally pollaxed me. I am due for my third shot next week. The week I had the N&V, I swore I would not have any further treatment, but no have the dilema of f F 48 2 months

5  premenopausal breast cancer After 2 yrs in combination with tamoxifen: thin eyebrows, cramping in calves, no periods (great!), but finally some severe cramping in calves and pain/swelling in am in knuckles and soles of feet. But I would do this combo again - not bad at all. Effects may be just menopausal. Used Lupron with Tamoxifen for 2 years for hormonal treatment of breast cancer. I believe Tamoxifen does stimulate the endometrium because I got my period back after chemo, 1 month after starting tamoxifen. Therefore I used Lupron for ovarian suppression to hopefully keep my estrogen down and prevent reoccurance of my breast cancer. I think it was a good decision - no cancer return so far, and quality of life was OK. F 47 21 months

3  Pre surgery for ablation Extreme hot flashes and night sweets and headaches. Recently severe chills, don't know if it related. The cold chills started about 2 weeks ago (just after my 2nd shot) and last about 30mins to an hr, until a hot flash takes over. I am painfully cold and can't get warm no matter what I do. The headaches tend to be mostly at night as well. I also had some leg pain the fist 2 weeks but it went away so I don't know if it was related either. I have one more treatment of Lupron and then I will be very glad to be done with it. The hot flashes and night sweets are causing a major lack of sleep and is wearing me down to the point where it is getting hard to cope. At night there are more like an attack. I also have type 1 diabetes and have a hard time telling a diabetic low from a hot flash which can cause, some serious problems. I only have to have the third shot as my surgery date has been moved. I don't think I can stand to have a 4th one if they move my date again. Two to three months of the side effects will hopefully be worth it for the surgery to be effective.  1  Adenomyosis I've suffered with adenomyosis, probably for 35 years. After years of being on various low-dose types of the Pill and Depo Provera, nothing was very effective. With Lupron Depot, I had hot flashes, cold chills, and sweating all day. I since decided to have a hysterectomy. I developed pain in my neck, arm/leg joints, and hips five months after my first (and last) injection. The joint pain has been getting progressively worse. I did not have joint pain prior to taking this drug. The 11.25 dosage is to be taken every three months. Four months after the injection, I had a hysterectomy (with my ovaries remaining). I'm not in menapause and am still ovulating. Prior to surgery, I was in excellent health, with no family history of arthitis. I am very strong and was very mobile prior to surgery. I'm taking it easy, as it has only been five weeks since my surgery, but I walk and move around and eat and sleep well. But I'm surprised that the neck and joint pain started after the surgery (five months after the Lupron Depot injection). The hot flashes and sweats have been greatly reduced since I started taking sage leaf extract a couple of times a day. But I'm concerned that the drug has affected my immune system, and I hope it hasn't triggered an auto-immune response in my body. F 47 3 months
