PROMETRIUM  (PROGESTERONE):  Progesterone is a type of female hormone (progestin). This medication is similar to the progesterone that your body naturally makes and is given to replace the hormone when your body is not making enough of it. In women who are not pregnant and not going through menopause, this medication is used to restore normal menstrual periods that have stopped for several months (amenorrhea). Progesterone is also used as part of combination hormone replacement therapy with estrogens to reduce menopause symptoms (e.g., hot flashes). Progesterone is added to estrogen replacement therapy to reduce the risk of cancer of the uterus. This medication is not for use in children. Progesterone must not be used to test for pregnancy.   FDA Approval Date: 1999-10-15 (Sources: U.S. Centers for Medicare Services, FDA)

5  to balance the estrogen I take I just reduced the progesterone again, to just 25 mg/night. After a month on the 50 mg dose I began to get depressed, tired and lethargic. So for me, 25 mg doesn't have that effect. F 53

4  premenstrual syndrome Fatigue: I take it at night one 100mg caplet, orally. Some fatigue but really not many side effects at all. Prometrium at the 100 mg dose has significantly improved my severe PMS. Take on days 11-25. I still have some mood changes during the premenstrual phase, but much better. 200mg was too much and I was moody and aggressive, so went back to the 100 mg. My periods start now without a week of spotting prior to, the flow is lighter and only last for 3-4 days instead of 7-10 days. My progesterone levels were low. I do not use any estrogen. I highly recommend prometrium for severe PMS. F 45

3  Luteal Phase Progesterone Increase Bloating, twinges in uterus, slight dizziness, pain down backs of legs as if they were bruised (especially my calf area), yellowish discharge, constipation. This is my first month trying Prometrium with IUI and 100mg Clomid. My progesterone level was 4 on CD 13. I haven't been tested to see if it is at a good level to sustain a pregnancy (although I am not sure if I am pregnant yet or not), but we'll see!! Although it is inconvenient (3x per day vaginally) I would continue taking this medication. F 27

Can't wait to get off this drug, but worth it if keeps preg stable F 37

3   Night sweats, fatigue, PMS symptoms. I use bioidentical estradiol and testosterone. Since I have my uterus I must also use progesterone to prevent endometrial cancer. I use Pometrium for 12 days, every second or third month. It causes night sweats, sleepiness and fatigue, bloating, and PMS symptoms. When the 12 days are over, I get a mild period and start feeling better quickly. It serves its purpose, but I don't love how it makes me feel. My doctor suggested inserting the capsules vaginally to reduce the symptoms (oral meds must be processed via the liver, creating metabolites which worsen side-effects). This does reduce the severity, but doesn't eliminate them. F 48

3  Menopause Awful menopause symptoms. Went on Oestrogen patch which really worked and after two weeks felt like my old self again. Had to introduce Progesterone and was prescribed Prometrium. LOVED it at first, slept like a log, first time in years and it was just fabulous with no side effects, Have been on it 2.5 months now and going off it as after about 6 weeks I gained so much weight and huge, distended, bloated stomach. I literally starved myself (always a slim person) but could not lose a single gram of weight. Huge boobs and just uncomfortable. Also, had a liver function test and my liver count was alarmingly high. I don't drink at all and the doc said "Progesterone' ... It has been one week now since I've been off it and already my stomach has gone down by 3/4 and my boobs too but it will take a while to get rid of this unusual fat that I have gained. Wonderful initially and wish it had stayed that way. Might try applying it occasionally and topically (by piercing capsule with a needle) in a few months time. F 54

3  Post menopausal symptoms If you have side effects, you may have high levels of progesterone due to drug-drug or drug-food interactions. DO NOT CONSUME GRAPEFRUIT PRODUCTS! The CYP3A4 enzyme clears most of prescribed drugs (substrates) from your system. Check to see if any of your meds or supplements are CYP3A4 substrates and or if they are inhibitors of CYP3A4. Grapefruit stops the action of CYP3A4 gut enzymes completely in 30 minutes, permanently and irreversibly. You renew the enzyme in over 24 hours, so taking grapefruit products separately from your drugs does not help. When you can not clear substrates, levels of them get too high and side effects, even fatalities, may occur. Many herbal supplements are CYP3A4 inhibitors as well. Do not take them. Ginkgo, garlic, turmeric, for example. Research every substance for CYP3A4 interactions. Be well! F 57

2  achieve/maintain pregnancy tender breasts, hunger, dizziness 1 hr after taking medicine, which lasted for about 1 hr. OB put me on Prometrium for low progesterone at week 5 of pregnancy. I started spotting at week 6, which became heavier and heavier until I miscarried after 8 weeks. My OB said to use Prometrium to extend Luteal Phase to acheive pregnancy after my miscarriage, but every month that I have taken it (4 months now) I start bleeding before I even have a chance to test for possible pregnancy. My OB says to just keep trying with the Prometrium, but I'm scared to take it now. It doesn't appear to be helping, and I'm begining to wonder it might actually be hurting our efforts to concieve.  2  menopause Tired, lethargic, depressed, can't wait to go home every day and just lie down. I had the worst dizzy spell last night. This has been going on for a month and prometrium is the reason. And the doc upped it to 200 mg which I bought and will have to throw away, as the pills cannot be cut in half. I don't think it matters what time of day I take it. I have the side effects 24/7. F 54

1  oral progesterones are not effectiv Progesterone works best when applied topically in the form or a USP progesterone cream. When applied in the suggested amount, which is about 1/4 tsp/day, it's the same amount the body makes. There are no bad side affects and the positive benefits are phenomonal--eases endometriosis, cramping, regulates periods, counter-balances estrogen and can help with fertility issues. F 51

4  Severe hot flashes - Horrible None as of yet, but.... was only up twice during the night for the first time in months. Only damp in morning instead of soaking wet and having had changed the bed during the nite. Maybe my days of sleeping on towels is over. This drug is like Plan F, they have changed me with all different combinations of drugs. Drugs seem to work for a little while then nothing. We'll see.... F 49 1 days

3  Induce period Light-headed!!! Hot flashes. I took this in the morning before going to work, in combination with estradiol. I thought the drugs would even each other out so that I wouldn't experience side effects, but no such luck. Based on the other comments I will start taking Prometrium before bed so that I can actually function at work. the last time I was on Prometrium I couldn't finish the 10 days I was so sick.  3  Post Hysterectomy I just started taking 100mg per night. I have been on Premarin for several years, however the Dr. feels I will benefit from having progesterone to balance out my HRT. Today is the first day and I feel very spaced out and emotional. Do you know if these side effect go away once my body adjusts? F 50 1 days

2  menopausal symptoms Increased anxiety, very dizzy a couple of nights after taking it, weight gain, extreme increase in appetite, migraine headaches, fluid retention, restless sleep, moodiness/depression like symptoms I actually was on this for approximately two months, ...then went off because of a constant headache over and behind right eye. Menopausal symptoms returned...decided to give it another try with the addition of migraine medication to alleviate the head pain. Worse side effects this time round. VERY dizzy after taking it, approximately 1 week into program. Can't seem to fall asleep very easily...cramps in the middle of both calves, wieght gain, which I am definitely not happy about, in that I am very much into fitness, and at barely 5 feet tall, I do not like the extra weight...very uncomfortable as there is also fluid retention in my legs. Would advise not to take this medication..will find another more natural way to alleviate my symptoms with menopause! F 46 1 days

2  Menoupause symptoms Racing heart, numerous hot flashes, insomnia, jittery feeling into the next morning. Tried this in combination with Divigel (estradiol) for mild menopause symptoms, the worst of which is insomnia. The very first night I took this I noticed side effects. It made my heart pound all night long and caused numerous hot flashes that woke me up every hour. During the night, I felt exausted, dizzy and loopy yet I could not sleep. I still felt jittery the next morning. Too afraid to feel that way another night, so it's going right in the trash. Too bad, because it's expensive! F 49 1 days
