LUPRON DEPOT  (LEUPROLIDE ACETATE):  Leuprolide is used to treat advanced prostate cancer in men. It is not a cure. Most types of prostate cancer need the male hormone testosterone to grow and spread. Leuprolide works by reducing the amount of testosterone that the body makes. This helps slow or stop the growth of cancer cells and helps relieve symptoms such as painful/difficult urination. Talk to your doctor about the risks and benefits of treatment.   FDA Approval Date: 1989-01-26 (Sources: U.S. Centers for Medicare Services, FDA)

1  Endometriosis Never ending side effects. I had this drug injected in me with hopes it would stop Endometriosis spreading. It caused me to eventually have to give up my career as a child psychologist to being totally disabled with Endometriosis growths and fibromyalgia, RA, muscle spasms and ptsd. It never leaves your system and attaches to your dna and changes it. I miscarried at 16 weeks after my last injection 4 months before I was pregnant. I’ve never been pregnant since then in 1995. I am now on stron pain meds just to function and spend many days in bed. Do not take this drug poison under any circumstances. It will ruin your health for life and not cure what the docs say. They get paid off to prescribe it by big pharma. Do not take this medicine!! It’s pure poison. F 49 6 months

1  shrink fibroids prior hysterectomy Horrible - blood pressure increased to 158/118 (normal for me 110/70). lightheadedness, dizzy, legs and arms swelled, extreme fatigue, jitteryness, shortness of breath and it never worked! Not worth it. I had one 11.25mg shot in Dec 09. Wish my doctor had told me the true side effects not just menopausal. Been to cardiologist, pulmonologist and endocrinologist since taking Lupron Depot and they are trying to fix the damage the drug has caused to my body. So far it has caused breathing problems (per pulmonologist), I have hypothyroidism and high and very low blood pressure. My lightheadness has never gone away. 5/09 I had a complete physical and was in perfect health. F 49 1 times

1  Endometriosis Serious permanent side effects. Memory loss, fibromyalgia, chronic fatique, huge weight gain, arthritus, herniated discs, neck damage, swelling/fluid retention, allergies so severe surgery was required and fluid drains out my ears all day, IBS, headaches, mania, suicidal depression, post traumatic stress, susceptible to catch germs-colds. These are all PERMANENT and I haven't had a shot of lupron for 8 years. If you want to gamble with your life then try Lupron, but I suggest your read the package insert first. I lost everything to this drug, my career, social life, I moved to try to improve my health. Here's what you need to know: Lupron causes a phenominon called F 49

1  Fibroids Horrible Muscle and Joint pain, doc now saying he believes i may have rhumetoid arthritis from taking the depot lupron. I never had bone or joint problems before, from a scale of 1-10 the pain everyday is a 8, i would tell anyone to not take depot lupron as it has torn my body up terrible and no shrinkage of the fibroid tumors. The pain i carry in my body now and headaches are horrific. I took two shots and each shot last three months, I wish i would have not taken this depot lupron F 49 2 months

5  Endometriosis, Tumors, Bleeding Increased hot flashes, chills, night sweats, mood swings. These symptoms had already begun before Lupron due to my premenopausal state but did increase. The bleeding stopped a couple of weeks after 1st (3 month)shot Aug05 I had been bleeding severely aprox. 300 days out of 365 the year or so before treatment. I completed 3rd shot Feb06 It is Jun06 now and still no period Yeay! Still have some cramping and lower back pain. No weight gain, acne, headaches or joint pain like others. I actually lost about 20lbs I discovered that exercise helps the hot flashes more than anything else other than a cold wet rag or a dip in cool water. Go figure. The exercise seems to regulate body temp the best ironically. Eat lots of ice-cream and chipped ice for bone and body health and cool down and you can make it without add back therapy. I hope I never have a period or cramps and vomiting again. I had endo so bad never could carry child to term also had tubal pregnancy and chronic pelvic inflammatory disease. Supposed to go back to Doc after period starts back to see if tumors and endo have shrunk 10 months have passed and still no period Yeay! F 48 10 months

4  Fibroid Tumor and Enlarged Uterus I am usually a very happy go lucky person. First injection caused crying outbursts, daily headaches and loss of appetite. I had the worst period that I have had since I was probably 16 years old. Acne around the forhead seems to be a problem although I have never had that problem before. Second injection I no longer have the headaches but I am still very emotional. I feel like someone zapped all my energy and my mid section seems to be getting thick although I am eating far less and have no appetite. I had a minor hot flash yesterday for the first time and do not look forward to anymore. On the positive side my back pain has all but disappeared. I no longer get up in the middle of the night 3-4 times to go to the bathroom. I can't remember the last time I slept through the night until I got my first injection. I don't have to wear a pad everyday anymore worrying about leakage. I know the drug is working. I just hope I can hang on for the third. I do feel sorry for my husband who doesnt know what he's comming home to each day! F 48 2 days

4  large fibroid; critical hemoglobin fatigue After reading these, I'm still waiting for the 'other shoe to drop' and find myself growing a third ear :). Caution is the key, but my experience has been no side effects yet except fatigue, which I cannot preclude from being from the critically low hemoglobin. I was given the 3-month dose (11.25). My goal is to NOT have a hysterectomy but only fibroid removal. It is too large currently for myomectomy alone, plus I would have to be transfused prior to having surgery. My blood loss definitely stopped (at least to this date.) It's early in treatment, so I can report more thoroughly at a later date. F 48 2 weeks

4  one shot prior to surgery hot flashes, weight gain, diminished sexual desire, unable to sleep, My doctor said this would minimize my chances of bleeding, stop periods, set my uterus in a better way for him to perform laproscopic surgery for its removal. I had one shot in May 05 and surgery was done Aug 05. when does this shot wear-off? Still having hot flashes which disrupt my sleep. They prescribed Premarin and when I read all the side effects it has I didn't take. This was a preview of what true menopause is going to be like-what a nightmare time for a woman. I just want to get back to feeling as i did before the shot which was actually really great accept for having a period every other week that last for 5-6 days. I had fybroids but not painful yet. Anyone know when the side effects will go away? F 48 0 days

3  Large Uterine Fibroid, anemia Minor bleeding and mild headache. I have not really noticed any symptoms but it has been less than one week since receiving the first injection. I am hoping for decrease in the size of fibroid to avoid surgery or at least postpone it for a while. Minor bleeding started 4 days after receiving the injection on 6/8/207. I will report back with an update prior to next injection. F 48 6 days

3  Oestrogen sensitive Breast Cancer Vaginal dryness - total atrophy, especially the first time I took it. Nothing helped. Within a month, sex was impossible. Hot flushes, reduced concentration and brain-power (I am a full-time student of Pharmacy). Exhaustion, sleepiness, joint pains. Probably caused the osteoarthritis detected in my recent scan. Osteoporosis developed very quickly, but bones improved quickly when I went off the drug for a while. I now take Actonel (a bisphosphonate) to prevent bone loss. I developed metastases when I went off the drug for a while, including while I was in a gene therapy trial. Don't people in the States take this drug for breast cancer ? It really does work if your cancer is oestrogen sensitive - you take it if you are pre-menopausal, to shut down oestrogen production by the ovaries. Then you can - and probably need to - take Arimidex as well, to stop peripheral production of oestrogen (by the fat cells, adrenal glands, etc.) I need to take both, as the Prostap / Lupron alone doesn't totally control the cancer. By the way, in the UK, Lupron is called Prostap. I have been living with breast cancer since at least 1997, and have only had this anti-oestrogen therapy - not chemo, because I had hoped to have children and didn't want to damage my fertility. Have taken Lupron / Prostap on and off for periods of 1-2 years. F 48

2  adenomyosis and fibroids just very tender at injection site for several days I took the shot on March 1. I was diagnosed with adenomyosis and fibroids. I was hoping to have tumor ablation but the only treatment for adenomyosis is to remove the uterus. I am going in for a supracervical hysterectomy in June. We can't do it any sooner since I am anemic and the fibroids are large. The lupron shot was to shut down my ovaries so that I would stop bleeding and build my blood up and hopefully shrink the fibroids. I have always had heavy periods and they have gotten worse over the last few years. On the 22 day into my cycle I started bleeding, by day 2 I was flooding every hour changing a super plus tampon and over night pad. I am on my third day and it is finally letting up some. Hopefully I will stop soon and not bleed the next two months. I need to be stronger for surgery. I know spotting was a possibility but this was awful. I started taking vitamin k and vitamin d drops. Will let you know how it progresses. It was an awful expensive mistake if it doesn't work. Thank God my insurance paid for it. Other than that I can't say that I notice any side effects. Hope it works better for others. I was so looking forward to giving the rest of my tampons to my daughter. I ended up having to go buy more. F 48

1  Endometriosis Taken in 2000, yes that is 2000 and Lupron has ruined my life and left me disabled permanently. I suffered immediate toxic reaction side effects that have destroyed my immune system. Doctors at the Medical Hospital of Virginia - MCV advise the med altered my immune system and physiologically changed my body causing MULTIPLE AUTOIIMUNE DISEASES THAT HAVE ALMOST COST ME MY LIFE REPEATEDLY. RUN, DON'T EVEN CONSIDER TAKING THIS TOXIC SHAM OF A DRUG THAT CONTINUES TO RUIN LIVES AND KILL ITS VICTIMS!! FORCED TO HAVE RADICAL HYSTERECTOMY, RHEUMATOID ARTHRITIS, LUPUS, CATARACTS, SJOGREN'S DISEASE AND ON AND ON I WAS 37 WHEN I TOOK LUPRON 11 YEARS AGO IN 2000 F 48 6 months

5  Endometreosis Lowered Libido, Hot Flashes, Tiredness, Increased Anxiety Spoke with my Dr. about side effects - she prescribed the Vivelle Dot 2.5 patch (change twice a week). Worked wonders. My GP prescribed anti-anxiety to help with the anxiety and tiredness. Both those meds made the world of difference. I have not known until this past year what "normal" feels like. I lived with pain so long, I thought that much pain was normal. I hope that I continue period/pain free since my last shot was in December. I guess I'll have to wait and see. I fought taking Lupron until I realized that I was ready to give up pain and periods. THANK GOD FOR LUPRON! F 47 12 months

4  endometriosis so far - slight headache, slight breast tenderness, slight tingle of skin haven't been on it long enough to know if it is working for me. I did notice a relief from my cramps right away - but don't know enough about the drug to know if it is actually the drug or my mind making the pain less. I have been in a really great mood ever since recieving my first shot - we have all been joking that maybe that is my mood change? would appreciate any comments as to how soon this is actually supposed to take effect.  4  Fibroid Causing Urinary Retention The first few weeks I experienced some nausau, but not too bad. Also some water retention in my ankles. I may have a slight increase in incidents of migraines but hard to tell. The shots are not painful, although for a day or two I tend to have a sore muscle at the injection site. At week 5/6 I started to notice that shrinking was happening as my urine flow was much better and I wasn't experiencing retention issues that I have had for four years. I have had only one instance of breakthrough bleeding, but very light. I have now had 3 monthly shots and find this drug quite tolerable. Have night sweats, but already had those anyway, and a bit more hot flashes. Doctor has advised me that maximum shrinkage occurs between 3 and 6 months. Also that you can remain on this drug longer than 6 months with therapy to ensure no loss of bone density. This drug will not cure me, but is allowing me time to shrink the fibroid and make it softer for me to consider mimimally invasive hysterectomy instead of abdominal surgery. That makes this drug worth trying. If you are concerned, ask your doctor for the one-month dosage and not the three-month. I have already had the Uterine Artery Embolization twice. No one ever told me that 20% of women will experience the fibroid generating alternative blood supplies allowing it to continue to grow. So, four years after starting to be catheterized to urinate I am still dealing with this issue. Just realize that after Lupron the Fibroid will grow back unless you have made it over the magic menopause cycle. So for me that means I will need to have surgical intervention once the Fibroid has shrunk. F 47 8 weeks

3  Endo 4 hot flashes/night sweats I took three doses of Lupron (1 per month), as recommended by my doctor. Had the needle in my hip - no pain at all. After I got the hot flashes/night sweats under control with the add back therapy, it was great. (those night sweats made life miserable - no sleep) Only problem...after five months, my period came back - still causing pain. My 'freedom' was short lived. F 47 3 months
