PROLIA  (DENOSUMAB):  Denosumab is used to treat bone loss (osteoporosis) in women who are at high risk for bone fracture after menopause. It is also used to treat bone loss in men who are at high risk for bone fracture. Osteoporosis causes bones to become thinner and break more easily. Your chance of developing osteoporosis increases after menopause (in women), as you age, if someone in your family has osteoporosis, or if you take corticosteroid medications (such as prednisone) for long periods. Denosumab is also used to treat bone loss in women who are at high risk for bone fracture while receiving certain treatments for breast cancer. Denosumab is also used to treat bone loss in men who are at high risk of bone fracture while receiving certain treatments for prostate cancer that has not spread to other parts of the body. This medication works by slowing bone loss to help maintain strong bones and reduce the risk of broken bones (fractures). Denosumab belongs to a class of drugs called monoclonal antibodies. It prevents certain cells in the body (osteoclasts) from breaking down bone.    (Sources: U.S. Centers for Medicare Services, FDA)

1  Osteoporosis Worsening back pain, nausea, bone pain in legs that come and go and joint pain. The bone pain was sharp, burning and shooting pain while sitting at my office desk. Never had that happen until after the injection. Also having severe muscle spasms especially in arms and back! Do not plan on taking second injection. Would rather take my chances with the osteoporosis!! 11 days

1  Osteoporosis diagnosed 17 years ago Swelling of ankles and feet, pain all over body as if bruised, cramps in feet, feeling confused, general tired feeling and lethargy. Started feeling poorly after 3 weeks of the shot. I have taken all the other meds over the years and this was my last chance. I really hate to decide between this terrible situation I now experience or the chances of a broken bone. 66 5 weeks

1  osteoporis I take prolia shots 2 x yearly. Prior to starting prolia my carotid arteries in 2008 were clogged at 35% left carotid and 30% for the right. Now an arteriogram shows both severely clogged with my left carotid at 80%, surgery is scheduled 19 Feb 2016. Taking the prolia is the only lifestyle change that occurred. Could Prolia have contributed to the carotid artery stenosis. I have taken 7 prolia shots and in March 2016 will take the next one if I decide to continue. I would be interested to know if anyone else has the clogged arteries after taking prolia for a while. 73

1  Osteoperosis First 3 injections were OK. I had a 6 month break and then a 4th injection. Within two weeks I could hardly move. Felt like all my leg, grion and arm muscles had been kicked badly. 5 weeks later my Physician took blood and diagnosed very severe calcium deficiency and extremely high Thyroid levels. After urgent treatment and 6 weeks later my levels are back to normal, but pain still exists and is worse in the first half of the day. I am male, and 72 with only 30% kidney function. I live in Australia. I will never tough Prolia again, and worry about how it has ruined my quality of live. I was a very active person, and now have to get gardeners in etc do work I now cannot even think about. It has wrecked my life. 72 4 years

5  Osteoporosis After 1 year, only 2 shots, I still had osteoporosis. I was encouraged by my dr to continue the shots for 1 more year. After 2 years on prolia my bone density now showed osteopenia! I never had a negative side effect! But something very positive did occur totally unrelated to osteopenia and I believe but have no proof that the prolia had totally taken away the extreme pain and fluid development I had been experiencing in my right knee for the past 5 years diagnosed as bone on bone! Can anyone explain this to me? F 71 2 years

5  Previous hip fracture Only side effect so far is a bladder infection. No pain, no rash, no jaw problems. Next injection is January 2014. The injection was completely painless. I also enrolled in the Prolia Patient Support Program and now get periodic emails with information on exercises and resources. I also received two telephone calls letting me know that I might be eligible for free injections. But my insurance company pays 100% of the cost so I paid little attention to the information.  5  Osteoporosis Dosage: 2 sub-Q shots per year. I had one shot 2 months ago and haven't had any side effects at all, thank God! I hope it continues to be side-effect free in the future. I am almost 51 and I have osteoporosis. I broke my ankle a few years ago just by rolling it off a sidewalk. It was very painful with a long recovery. This medication is supposed to increase bone density. There are also risks for one's blood calcium to decline while taking this medication. F 50 1 times
