PROLIA  (DENOSUMAB):  Denosumab is used to treat bone loss (osteoporosis) in women who are at high risk for bone fracture after menopause. It is also used to treat bone loss in men who are at high risk for bone fracture. Osteoporosis causes bones to become thinner and break more easily. Your chance of developing osteoporosis increases after menopause (in women), as you age, if someone in your family has osteoporosis, or if you take corticosteroid medications (such as prednisone) for long periods. Denosumab is also used to treat bone loss in women who are at high risk for bone fracture while receiving certain treatments for breast cancer. Denosumab is also used to treat bone loss in men who are at high risk of bone fracture while receiving certain treatments for prostate cancer that has not spread to other parts of the body. This medication works by slowing bone loss to help maintain strong bones and reduce the risk of broken bones (fractures). Denosumab belongs to a class of drugs called monoclonal antibodies. It prevents certain cells in the body (osteoclasts) from breaking down bone.    (Sources: U.S. Centers for Medicare Services, FDA)

1  osteoporosis I have chronic pancreitis, and this stuff has REALLY made my pancreas hurt. It is swollen as is my whole belly. Plus, at three months my hair began to fall out and is is totally ruined now. I have approximately 1/2 left and what is left is porous and straw-like. I have also experienced very high anxiety. If you have other health problems, I do not recommend you take this drug. This drug actually changes your hormones and 'shuts down' the body's natural sloughing of bone cells. The idea is to stop the sloughing, but not the making of new cells. Anything that works in this manner has got to affect other hormones. I'm scared of it. I will NOT do the second shot. The choice is brittle bones and my sanity or stronger bones...maybe....and I die of pacnreatitis or otheer problems. No thank you. F 5 months

1  Osteoporosis Worsening back pain, nausea, bone pain in legs that come and go and joint pain. The bone pain was sharp, burning and shooting pain while sitting at my office desk. Never had that happen until after the injection. Also having severe muscle spasms especially in arms and back! Do not plan on taking second injection. Would rather take my chances with the osteoporosis!! 11 days

3  Senile Osteoporosis These injections HURT!! Then it burns for a few days at injection site. They can't be given in the muscle, but in the arm area where most people have some fat. I don't really have any fat, and it's really painful to get. Then the injection site burns for a few days afterwards. Other than THAT, knock on wood, I have not had any other side effects that I'm aware of. I've had 3 shots so far, get my 4th in July and a follow up bone scan next January to see if it's working for me. F 46

1  Osteoporosis Agonizing pain in my neck, back, ribs, and my joints are all swollen and painful. Increased migraines, gas and constipation, bladder and urinary pain . It actually hurts to breathe! This was my 2nd injection and I hoped for a different experience, this time. 3 weeks later, I am in agony!! Last time the agony lasted 3 1/2 months. some days I just cried, the pain was so bad. I made a commitment to take Prolia because my osteoporosis is so bad, and my G.I. system can't handle any of the other drugs. I don't know if I can keep that commitment.. I don't know if I would have made the commitment to this treatment, if I knew all of the misery it would be. I have just about every side effect listed, but my doctor and I agreed this is the only drug that will work for my very severe osteoporosis. 1 more year 2 injections to go, before I get to take a, "drug holiday." If I survive that long! F 49 7 months

5  Osteoporosis Dosage: 2 sub-Q shots per year. I had one shot 2 months ago and haven't had any side effects at all, thank God! I hope it continues to be side-effect free in the future. I am almost 51 and I have osteoporosis. I broke my ankle a few years ago just by rolling it off a sidewalk. It was very painful with a long recovery. This medication is supposed to increase bone density. There are also risks for one's blood calcium to decline while taking this medication. F 50 1 times

1  Osteopina The FDA and Amgen should be ashamed. This drug either should not have been approved or mandatory information given to the patient. This drug has sent me to the emergency room for shortness of breath, heart palpitations, headache, blurry vision, severe muscle and bone pain. I have been sick for a year. I sometimes feel like I am actually dying. I have one month left of this injection left in my body and I pray no permanent damage has been done. I belong to a group on Facebook about Prolia complaints and there are hundreds of sick people because of Prolia. I realize some life saving drugs can be harsh but osteopina is not. Seems like there is a rat race with the pharmaceutical companies to hurry and get their drugs approved but without proper investigations and studies. Never, ever again will I be poisoned by Prolia. I would rather break every bone in my body. If you are thinking of getting Prolia injections investigate and study and learn about the drug. Once you are injected and if you have side effects you will suffer for at least six months. F 50 1 years

1  Ostopina Just had my 4th shot, 1 every 6 months. My oncologist down played the side effects. I have an infected tooth and my oncologist said the side of effect of ONJ, the bone comimg out of the jaw so that it is exposed, is too high of a risk to get the tooth removed. This drug makes it so the bones in your jaws dissenergrate and the gum doesn't heal and the bone in your will become visible and infected. (Google it). The doctors must be getting a kick back from Amgen when they prescribe it. At first I did fine now I don't want to walk, play tennis or do fun things with my kids because I feel like I have the flu and my right arm hurts. When I wake up in the morning my teeth feel akward. Two years ago I read the side effects in the literature from Amgen and ONJ -jaw rot was a rare side effect now Amgen says it is a common side effect. Stay away from this drug...after 2 years you will be lethargic, 15 pounds heavier, your teeth will feel loose, your skin will be flaky, your bones will ache. I take the calcium supplements with vit D too. Don't let Amgen say your are having these side effects because you're not taking calcium. Finds other ways to get your bones strong. Amgen just wants money..this drug is very expensive. The side effect of ONJ is horrible! Shame on Amgen for creating this drug and the doctors for prescribing it. I hope the CFO and the President of Amgen along with my oncologist get ONJ l and wake up every day feeling like their teeth are going to fall out. F 52 2 years
